Design Therapeutics announced the initiation of patient dosing in its Phase 1 multiple-ascending dose (MAD) trial for DT-818, a GeneTAC® small molecule targeting Myotonic Dystrophy Type-1 (DM1). The open-label study will assess safety, tolerability, pharmacokinetics, and pharmacodynamic effects, with data anticipated in 2027.
DM1 is a progressive neuromuscular disease affecting over 70,000 people in the U.S., caused by a DMPK gene mutation. Patients experience muscle weakness, cardiac arrhythmias, and neuropsychological changes. DT-818 aims to address the root cause by selectively reducing transcription of the mutant allele.
Key milestones include 2027 Phase 1 data readout and progression to later-stage trials. Competitive differentiation will hinge on DT-818's ability to demonstrate superior pharmacodynamics versus RNA-targeting approaches in development.
